The Reduction and Oxidation of Glutathione by Plant Mitochondria.

چکیده
is catalyzed by glutathione reductase (3, 4, 5). In earlier studies this reaction has been coupled with soluble dehydrogenases which utilize TPN as a coenzyme (6). Since tissue fractionation studies have shown that numerous dehydrogenases, including those associated with the oxidation of the Krebs cycle intermediates, are localized within the mitochondrion, it was of interest to determine whether these enzymes might also be coupled with reaction 1. This paper reports that it is possible to demonstrate the reduction of GSSG by plant mitochondria in the presence of suitable substrates. In addition, the plant particles also catalyzed the oxidation of GSH by molecular 02 in the presence of catalytic amounts of ascorbic acid.

A specific form of thioredoxin h occurs in plant mitochondria and regulates the alternative oxidase.
The plant mitochondrial thioredoxin (Trx) system has been described as containing an NADPH-dependent Trx reductase and Trx o. In addition to the mitochondrial isoform, Trx o, plants are known to contain several chloroplastic Trx isoforms and the cytosolic Trx h isoforms. We report here the presence in plant mitochondria of a Trx isoform (PtTrxh2) belonging to the Trx h group. Western blot analy...
متن کاملMitochondrial Toxicity of Depleted Uranium: Protection by Beta-Glucan
Considerable evidence suggests that mitochondrial dysfunction contributes to the toxicity of uranyl acetate (UA), a soluble salt of depleted uranium (DU). We examined the ability of the two antioxidants, beta-glucan and butylated hydroxyl toluene (BHT), to prevent UA-induced mitochondrial dysfunction using rat-isolated kidney mitochondria. Beta-glucan (150 nM) and BHT (20 nM) attenuated UA-indu...

متن کاملA comparative study of plant and animal mitochondria exposed to paraquat reveals that hydrogen peroxide is not related to the observed toxicity.
Rat liver mitochondria are much more susceptible to protein oxidation induced by paraquat than plant mitochondria. The unsaturated index and the peroxidizability index are higher in rat than in potato tuber. The levels of superoxide dismutase and glutathione reductase are concurrent with the different sensitivities to paraquat, with higher activities in plant mitochondria. However, glutathione ...
متن کاملThe metabolism of menadione impairs the ability of rat liver mitochondria to take up and retain calcium.
The ability of mitochondria to take up and retain Ca2+, and thereby to effect the free intracellular concentration of this ion, is well established. More recently, it has been reported (Lehninger, A. L., Vercesi, A., and Bababunmi, E. A. (1978) Proc. Natl. Acad. Sci. U. S. A. 75, 1690-1696) that the redox state of pyridine nucleotides modulates mitochondrial Ca2+ balance, since the oxidation of...
